Cell Counting Guide

Sep 04, 2026

Principle: Place the specially made cover glass on the counting chamber. A 0.1mm thick area is formed between the counting area and the cover glass. The counting area is 3.0mm x 3.0mm and is divided into 3x3 large grids. Each large grid is divided into 4x4 small grids (A, C, G, I zones). In zone E, each small grid is divided into 4x4 smaller grids. Therefore, the volume of each large grid is 0.1 x 1.0 x 1.0 = 0.1mm³.

① Clean the counting plate thoroughly, then dry it. Place the cover glass on top.
② Prepare the cells into a single-cell suspension and drop it onto the counting plate.
③ Wait for a moment and then count under the microscope.
Method 1: Count the cell density in zones A, C, G, and I.
(A + C + G + I) / 4 x 10^4 cells/mL
Method 2: Count the five small grids in the upper left, upper right, lower left, and lower right of zone E.
Cell density: Total number x 5 x 10^4 cells/mL
