How To Use Petri Dish

Petri dish is a kind of laboratory utensils used for microorganism or cell culture. It consists of a flat disc-shaped bottom and a lid, and is generally made of glass or plastic. The materials of petri dishes are basically divided into two categories, mainly plastic and glass. Glass can be used for plant materials, microbial culture, and adherent culture of animal cells. The plastic may be made of polyethylene, and there are disposable and multiple-use ones, which are suitable for laboratory inoculation, streaking, and bacterial isolation operations, and can be used for the cultivation of plant materials.
Generally, there are four steps of soaking, brushing, pickling, and cleaning.
1. Soaking: New or used glassware should be soaked in clean water first to soften and dissolve the attachments. New glassware should be briefly washed with tap water before use, and then soaked in 5% hydrochloric acid overnight; used glassware is often accompanied by a large amount of protein and grease, and it is not easy to brush off after drying, so it should be immediately immersed in clean water for scrubbing after use .
2. Scrubbing: Put the soaked glassware in detergent water and scrub repeatedly with a soft brush. Do not leave dead corners, and prevent damage to the surface finish of the utensils. Wash the cleaned glassware, dry it, and prepare it for pickling.
3. Pickling: Pickling is the process of immersing the above-mentioned utensils in a cleaning solution, also known as acid solution, which removes possible residual substances on the surface of the utensils through the strong oxidation of the acid solution. Pickling should not be less than six hours, usually overnight or longer. Be careful when handling utensils.
4. Rinsing: The utensils after brushing and pickling must be fully rinsed with water. Whether the utensils are rinsed clean after pickling directly affects the success or failure of cell culture. Manually wash the soaked utensils. Each utensil must be repeatedly "filled-emptied" for at least 15 times, and finally immersed in re-distilled water for 2-3 times, dried or dried and packaged for later use.
